This video demonstrates the loading sequence for running immobilized pH gradient (IPG) strips in the PROTEAN i12 system for isoelectric focusing (IEF) in the first dimension of two-dimensional (2-D) protein electrophoresis. The quick, easy, and foolproof loading and assembly of the focusing tray is illustrated step by step for the gel-side-down configuration; disassembly is also shown. The PROTEAN i12 IEF system can run up to 12 IPG strips from 7–24 cm long. Each channel in the i12 focusing tray is independently powered, giving the user total control over each IPG strip.
