Extraction of DNA from FFPE Tissue and Application in NGS

  • The analysis with Next Generation Sequencing (NGS) of DNA isolated from Formalin Fixated Paraffin Embedded (FFPE) Tissue is often a problem because this DNA is fragmented and cross-linked. To improve the success-rate with FFPE-samples, we compared our conventional FFPE-DNA isolation method with magnetic beads with a new method for FFPE-DNA isolation using the Covaris instrument. Our data of our first experiences with these procedures are presented. The Covaris method for FFPE-DNA isolation is an improvement compared to the method with magnetic beads. However, it also appears that the special sample prep methods for small quantities of DNA are anyway an improvement and promising for future experiments.
