  • "For more info, visit http://www.bio-rad.com/yt/16/proteonwebinar. Surface plasmon resonance (SPR) is known for its ability to obtain detailed information on intermolecular binding constants. However, the ProteOn SPR system can go beyond that and provide new and convenient ways to design various biochemical assays. We exploited it extensively to characterize the interaction of Diphtheria toxin with human antibodies against it. The ProteOn 6×6 array proved invaluable to map the antibody binding sites through competition experiments, showing that the most effective antibodies bind to the same region of the toxin. Then we used SPR to investigate the ability of the antibodies to inhibit binding of the toxin to its cellular receptor and to lipid membranes to better understand the virulence mechanism of this important human pathogen.
