Monitor 3D spheroid growth and shrinkage in real-time using IncuCyte™

  • 3D tumor spheroids were created by seeding A549 human lung adenocarcinoma epithelial cells, labelled using the NucLight Red reagent (Essen BioScience - Cat No 4476), into Ultra Low Attachment Round Bottom Plates. Spheroid growth was visualized and measured over a period of 10 days within a standard cell culture incubator using the IncuCyte ZOOM® live cell imaging platform. Automated image analysis by IncuCyte ZOOM® provides spheroid area and fluorescence intensity metrics. Spheroid treatment with the protein kinase inhibitor staurosporine (1µM) causes shrinkage of the microtissue and a reduction in fluoresce intensity compared to vehicle control.
