A look back at the second season of Behind the Science... 

Glyphosate is a commonly-used herbicide that’s a tough analytical challenge for food testing laboratories. Structurally, glyphosate is a small, polar compound that doesn’t have a chromophore so it can’t be detected by UV. Current analytical methods use labeling and GC-MS, but researchers would prefer to test for its presence against maximum residue limits (MRLs) directly. Jen talks with Jeremy Shia of Waters, who discusses an updated LC-MS method using HILIC chromatography with a Torus DEA column and quantification on the Xevo TQ-XS mass spectrometer. While phthalates may not be listed in ingredients, the concern is that they could pass as a leachable from packaging for cosmetics or personal care products, or from a contact material in beverage production into a bottle. In fact, for whisky import to China, a certificate of analysis is required for phthalates. The problem is that screening by GC/MS can result in false positives. Every good rock band starts in the basement. So that's where we're heading ... to the basement of Waters to see the first chromatography systems! Pat McDonald, corporate fellow emeritus at Waters, tells us about Jim Waters and Waters Associates' inventions. The first commercial "high pressure" chromatography system for polymer chemists, the GPC-100, from 1963. The ALC-100, developed in late 1970 for separating isomers in the synthesis of B12. The M6000 pump that came in 1972, followed by the U6K injector, the first commercial "small" particle columns (10 µm!). Bringing those pieces into an assembly led to the first modern "high performance" liquid chromatography instrument, now known as HPLC.
