In this webinar, Dr. Dominic Eisinger describes the development and validation of a multiplex biomarker panel for quantifying the commitment, onset, and induction of apoptosis by the intrinsic pathway in tumor tissue samples. 

Fourteen bead-based immunoassays for apoptosis-associated proteins were developed on the Myriad RBM multi-analyte profile (MAP) platform, which is based on Luminex xMAP technology. All assays were confirmed by IP-western blotting with the same antibody pairs used in the bead-based sandwich assays. 

The assays were manufactured as three panels: 
Panel 1: BAK, BAX, lamin B (intact +45 kDa), and SMAC
Panel 2: BAD, BAX/BCL-2 heterodimer, BCL-xl, BIM, and Mcl-1
Panel 3: Active caspase-3, BCL-xl/BAK heterodimer, MCL-1/BAK heterodimer, pBAD, and survivin 

Each assay was validated for precision, linearity, and accuracy for use with tumor biopsies, cell lysates, and tissue lysates. The quantitative assays were found to be more sensitive than IP-westerns.

Learn how researchers at the Immaculate Dermopathic Institute in Rome, Italy use the Bio-Plex system to help identify early markers of melanoma in serum and blood samples from at-risk patients. In studying the molecular mechanisms of growth and inhibition factors in this most deadly form of skin cancer, scientists in the Tissue Engineering and Cutaneous Physiology Laboratory hope to develop more effective therapeutics. 

The Bio-Plex system allows them to perform high-throughput multiplexed investigations of protein phosphorylation states, a key to the activity of many growth factors. Compared with the classical proteomics approaches, the Bio-Plex system significantly reduces the cost, time, and expertise required to perform these experiments. The Bio-Plex system allows for improved reproducibility and sensitivity with very small sample volumes, allowing the design of experiments that are not possible with current ELISA techniques.

The work in Prof. Facchiano's group will enable the development of new treatments to reduce the mortality rate of melanoma and also provide novel methods for the early diagnosis of this disease; both advances will ultimately save human lives.

Dr Richard P. Kraig presents a webinar on the use of cytokine analysis in the investigation of normal brain function. Dr Kraig and his colleagues use the Bio-Plex® suspension array system to explore innate cytokine signaling activity in the brain, focusing on downstream gene activation and protein transcription and how the resulting phenotypes affect neural activity. They hope to better understand intrinsic neuroprotection mechanisms, with implications for the prevention and treatment of neurological diseases such as epilepsy and dementia.

Dr Kraig discusses some of the issues encountered during work in his laboratory using femtomolar-level cytokine analysis in a murine model to study how environmental factors influence learning and neuroprotection, and to determine the cellular and molecular processes involved. The sensitivity, reproducibility, and high dynamic range of Bio-Plex cell signaling assays enable these researchers to obtain precise measurements of cytokine levels and gain insight into this biological system.
