Adeno-associated virus (AAV), with low immunogenicity and toxicity, has become one of the most promising viral vectors used for delivery of genetic material in gene therapy. Both are involved in the development of adeno-associated viral vectors, or AAVs. Ed works in upstream development, while Anne works on the downstream recovery and purification of biosynthetic products. 

00:25 AAV capsid titer is a critical quality attribute that must be monitored throughout process development and the production of AAVs, but Ed and Anne take two very different approaches.
00:38 Ed uses an ELISA, which requires six steps and takes three to four hours to perform. Unfortunately, Ed’s latest ELISA run delivered inconsistent results, so he’s going to need to stay late, again, and redo the ELISA.

00:38 BLI is an optical technique that analyzes the interference pattern of white light reflected from two surfaces: a layer of immobilized protein on a biosensor tip, and an internal reference layer.  Any change in the number of molecules bound to the biosensor tip causes a shift in the interference pattern, which is recorded in real time.

00:59 A proprietary protein coating at the biosensor tip acts as a binding partner, providing reproducible coupling of target molecules while minimizing non-specific binding. The measurement is plate-based, and aqueous buffer is used to generate a baseline, before the sensor dips into a sample containing the analyte. 

01:18 The analyte then binds to the sensor surface, and the binding interaction is followed in real time to reveal the association kinetics. The sensor tip is then dipped into fresh buffer, and the dissociation can be monitored until the regeneration of the biosensor surface. 

01:36 This real-time analysis provides precise and accurate data on binding specificities, analyte concentrations, and rates of association and dissociation.

01:48 A unique characteristic of BLI is that only analyte binding or dissociation can shift the interference pattern and generate a response profile. Unbound molecules, changes in the refractive index or pH, or other matrix effects do not affect results, making this technology well suited to measurements in crude samples. 

02:09 In addition, BLI is easy to use, making assay development and optimization a much faster process, for a shorter time to results. This fluidic-free approach eliminates sample loading or cleaning steps between measurements.

02:23 Finally, BLI is easily scalable. With the sensors arrayed in a standard SLAS-format, you can perform measurements with multiple sensor tips in parallel to perfectly match individual labs’ throughput needs.
