A total of seven new small molecule kinase inhibitors have been approved by FDA in 2017. Protein kinases form a very large family of signaling proteins, with more than 500 encoded in the human genome. They have many diverse roles in cells and can be regulated in a variety of ways providing essential nodes in intracellular signaling pathways that act to define and/or integrate intracellular communication. A protein kinase is a kinase enzyme that modifies other proteins by chemically adding phosphate groups to them. Phosphorylation usually results in a functional change of the target protein by changing enzyme activity, cellular location, or association with other proteins. Protein kinases fall into two major categories, serine/threonine-specific protein kinases and tyrosine-specific protein kinases. Antibody-drug Conjugates or ADCs consist of an antibody, a cytotoxic drug, and a linker that attaches the two. Such conjugates exhibit a broadly applicable method to increase the antitumor activity of antibodies and improve the tumor-to-normal tissue selectivity of chemotherapy. An ideal ADC has a selective monoclonal antibody and a highly potent drug, mixed with properly stable linker systems and conjugation technologies that preserve the characteristics of the antibody. Immunotherapy, a type of cancer treatment designed to eradicate disseminated cancer by harnessing the potential of the immune system. Immunotherapy agents do not directly attack the tumour but instead mobilize the immune system -this can be achieved through various approaches that utilize adaptive or innate immunity.

Historically, the mainstays of cancer treatment has largely relied on killing tumor cells with surgery,chemotherapy and radiotherapy. But these methods have limitations including severe systemic toxicities, bystander effects on normal cells, etc.

The critical relationship between immune function and cancer was first proposed by Rudolf Virchow 150 years ago when he observed the prevalence of leukocytes in tumours. For the next 100 years, there was little clinical demonstration that the immune system could be mobilized to provide a reproducible benefit for patients with cancer. Since the regulatory approval of ipilimumab in 2011, the field of cancer immunotherapy has been experiencing a renaissance. Immunotherapy has become an important part of treating some types of cancer owing to its unique science and its potential for substantial and long-term clinical benefit.
