By following changes in the fluorescence of the amino acid tryptophan or tyrosin, chemical and thermal stability is assessed in a truly label-free approach. The dual-UV technology by NanoTemper allows on-the-fly fluorescence detection, providing unmatched scanning speed and data point density. This results in ultra-high resolution of unfolding curves, which are not compromised by aggregation thus allowing detection of multiple, and even minute unfolding signals. Because no secondary reporter fluorophores are required, protein solutions can be analyzed independent of buffer compositions, with a maximal protein concentration range from more than 250 mg/ml down to 5 µg/ml. This allows for the analysis of detergent-solubilized membrane proteins as well as highly concentrated antibody formulations.
